At eighteen years old, Fatone hooked up with Lance Bass, JC Chasez, Chris Kirkpatrick, and Justin Timberlake to form the popular boy band, 'N Sync. The group has been credited with re-launching the popularity of the boy band genre and pop music in general, which had all but faded by the end of the 80s, following the dawn of alternative and grunge music.
‘N Sync released their last album in 2001, and have since been on hiatus, with the success of Justin Timberlake’s solo endeavors taking center stage. Since the group’s separation, Fatone has focused on his acting career, appearing on Broadway, in film, and television.
In 2002, Fatone landed the role of cousin "Angelo" in surprise-hit film My Big Fat Greek Wedding, starring Nia Vardalos. He has also appeared in films such as The Cooler in 2003, with William H Macy and Alec Baldwin; as well as Joe Dante's Matinee in 1993. In addition, Fatone has appeared along with fellow ‘N Sync star Lance Bass in Longshot in 2000 and On the Line in 2001. Bass and Fatone are currently collaborating on a reality show styled after The Odd Couple, which will feature the two former band mates paired as mismatched roommates.
In 2003 Fatone hosted talent competition Fame on NBC. Also on the small-screen, he has made an appearance on Seth Green’s Robot Chicken, being trained to battle Britney Spears; and on MTV Cribs, showing off one of his prized possessions – an actual "KITT" car used on the David Hasselhoff series Knight Rider.
On the stage, Fatone has appeared as Wolfie in Beetlejuice's GraveYard Revue, Seymore in Little Shop of Horrors, and as Mark in Rent.
In December 2006, Fatone told a Florida radio station that he’d like to take part in popular celeb-reality series Dancing with the Stars, even though he has turned down three previous invites to take part in the show. He competed in the fourth season of Dancing with the Stars and ended up coming in second place in the competition, after winner Apolo Anton Ohno. Also competeing in the fourth season were Laila Ali, Ian Ziering, Billy Ray Cyrus, John Ratzenberger, Heather Mills, Clyde Drexler, Leeza Gibbons, Shandi Finnessey, and Paulina Porzikova.
Fatone is married to long-time girlfriend Kelly Baldwin. They have one child together, daughter Brianna.
